Synovial inflammatory response to bacterial endotoxin.

چکیده
The ability of bacterial endotoxins to produce local inflammation has been recognized as one of the many biologic activities of these intriguing products of the gram-negative bacterial cell.1" This property of endotoxin, however, has not been studied extensively with the highly purified materials now available. Preliminary observations are reported here on the inflammatory response elicited by intra-synovial injections of endotoxin into rabbit suprapatellar bursae. These studies were designed to determine the sensitivity of this technique for detecting endotoxin contamination as a possible cause of the inflammatory reaction produced by intra-synovial injection of foreign proteins.' Because the synovial cavity proved extremely reactive to endotoxin-induced inflammation, the initial observations were expanded and compared with other known biologic properties of endotoxin. Repeated intravenous injections of endotoxin produce a relative resistance to many of its biologic effects, particularly the ability to produce fever and to cause death. This relative resistance has been termed 'tolerance' to endotoxin.' The inflammatory reaction in 'tolerant' rabbits was of particular interest.

Effects of drugs on different types of synovial inflammation in the rabbit.
Inflammatory joint diseases in man differ markedly in their response to anti-inflammatory agents. Perhaps the best known example is the dramatic effect of salicylates in acute rheumatic fever in contrast to their minimal value in acute juvenile rheumatoid arthritis. Phenylbutazone is quite helpful in treating ankylosing spondylitis, but is of little value in peripheral rheumatoid arthritis. In ...
متن کاملThe effect of bacterial products on synovial fibroblast function: hypermetabolic changes induced by endotoxin.
The effects of bacterial products on selected synovial fibroblast functions were studied. Extracts of commonly encountered microorganisms were prepared by sonic or mechanical disruption. "Purified" endotoxins were prepared from selected organisms, and in some cases were purchased commercially. Normal fibroblasts were derived from synovial connective tissue obtained from amputations or arthrotom...

Bacterial lipopolysaccharide (LPS; endotoxin) is a potent immunostimulant that can induce an acute inflammatory response comparable to a bacterial infection. Experimental observations demonstrate that this biological response can be either blunted (tolerance) or augmented (potentiation) with repeated administration of endotoxin. Both phenomena are of clinical relevance. We show that a four-dime...

The limulus assay for the detection of bacterial endotoxin has been applied to the study of synovial fluid. Three of 5 patients (60%) with culture-positive gonococcal arthritis had positive SF limulus assay results; as did 2 of 11 (18%) with presumptive evidence of gonococcal arthritis, 3 of 6 (50%) with nongonococcal infectious arthritis, and none of 47 patients with noninfectious arthritis. E...
متن کاملEndotoxin Neutralization as a Biomonitor for Inflammatory Bowel Disease
Gram-negative bacterial endotoxin is a potent immunostimulant implicated in the development and/or progression of a variety of diseases. The mammalian immune system has both innate and adaptive immune responses to neutralize endotoxin. In this study, a system was developed to monitor bacterial exposure by measuring the extent and nature of endotoxin neutralization in plasma. In control patients...
